THE STUDY OF DGAT1 AND PIT1 POLYMORPHISM IN ANIMALS OF THE ENDANGERED BROWN CARPATHIAN CATTLE BREED

The article presents the results of the study of structural genes in Brown Carpathian cows that are associated with productivity: DGAT1 (Diacylglycerol O-acyltransferase 1) and PIT1 (pituitary transcription factor). The study used 30 samples of DNA isolated from the venous blood of Brown Carpathian cows using the «DNA Sorb-B kit» (AmpliSens). Genotyping was performed using polymerase chain reaction (PCR-PCR) polymorphism analysis of restriction fragment lengths. As a result of the research, it was found that the polymorphism of the DGAT1 and PIT1 genes is represented by alleles DGAT1А, DGAT1К and PIT1А, PIT1В and, accordingly, by the genotypes DGAT1КА, DGAT1КК and PIT1АА, PIT1АВ, PIT1ВВ. For the DGAT1 gene, an amplified fragment of 411 bp. treated with CfrI restriction enzyme. A high frequency of the DGAT1К allele was established at 0.585 and a slightly lower frequency of the DGAT1A allele at 0.415. When studying the PIT1 gene, the amplification product (451 bp) was treated with the restriction enzyme HinfI. It was found that allele B (0.81) and homozygous genotype PIT1BB (0.67) were more common. The results of the study are valuable in connection with the sharp reduction in the number of small and aboriginal populations and the threat of extinction of the own genetic resources of agricultural species.
